4. Adamts (a disintegrin-like and metalloprotease with thrombospondin type 1 motif) expression in keloid and hypertrophic scars

Keloid is known as a proliferative and fibrotic lesion characterized by abnormal collagen deposition (collagen bundle) and excessive deposition of other extracellular matrix (ECM). It has been reported that excessive deposition of collagen and glycosaminoglycan (GAG) causes reduction in elastic fibers in keloid lesion. The role of ECM in keloid is still unclear. The aim of this study was to investigate versicanases expression in keloid. We investigated mRNA expression of versicanases (a disintegrin-like metalloprotease with thrombospondin type 1 motif: Adamts-1,4,5,9) in 5 cases of keloid tissue (2 cases with normal skin lesion), 3 cases of hypertrophic scar tissue and one normal skin tissue. RNA was extracted from the tissues and cDNA prepared by Reverse Transcription Polymerase Chain Reaction. RT-PCR. Real time quantitative PCR was performed to check the mRNA expression of versicanases. The results suggest that versican degradation by ADAMTS-4,5 may play an important role in keloid formation. Manipulating ADAMTS-4,5 or versican degradation may have therapeutic implications.
